I got a job in a museum not long after completing my MA in Museum Studies. I completed at least 20-25 applications in about 2 months towards the end of my course. I got two interviews close together and got one of them. Part of the reason I believe I got the job was that I had volunteered in a couple of museums (one full time for about three months), before I applied, and this gave me experience within the field. My volunteering jobs were a range of experience from collections, education to curitorial. This was useful when applying as I would be able to apply previous situations in the application/interview.

I started as a Gallery Attendant (semi-guard, semi-look like a cool kid and answer questions) for minimum wage directly out of my BFA. I volunteered on my two days off a week in several departments (Education, Rights & Repro, and Archives), eventually got a second part-time job working for Special Events at night. From my volunteer work I received a temporary job assisting the Head Archivist for several months working on a rather large exhibition we were loaning to. Finally I wound up Manager of Visitor Services when my former boss left the position.
From what I have been through and have seen in my young career (3 1/2 years so far) if you can show the desire and dedication along with a lot of hard work (and skill/knowledge of course) you can achieve almost anything.
